A novel duplex SYBR Green real-time PCR with melting curve analysis method for beef adulteration detection
2021, Food ChemistryCitation Excerpt :We also found that, the 50% bovine content nearly corresponded to the maximum intermediate peak area, and the 1% and 95% bovine content corresponded to the minimum intermediate peak area (Fig. 2c-h). The reason for this phenomenon was explicated that two or more mismatch universal products formed double strands DNA with fluorescent dyes, and then these tested fluorescent signals produced the intermediate peaks (Boyle, Dallaire, & MacKay, 2009; Pang, Cao, Zhang, & Lee, 2011; Waterfall, Eisenthal, & Cobb, 2002). The closer the total number of two kinds of universal amplicons was, the more likely they would form mismatched products, so the maximum intermediate peak area would be formed when two species had same universal PCR products.
Evaluation of different genomic regions of Rotavirus A for development of real time PCR
2019, Journal of Virological MethodsCitation Excerpt :The designing of NSP2 gene derived primers and probe described in a single study (Gautam et al., 2016) was based on the Rotarix vaccine strain, and hence, was useful only for determining the frequency of Rotarix vaccine strain and vaccine derived reassortant strains associated with acute gastroenteritis. On the other hand, the NSP3 gene based TaqMan assay has been reported widely for detection of RVA (Pang et al., 2004; Freeman et al., 2008; Jothikumar et al., 2009; Pang et al., 2011; Mijatovic-Rustempasic et al., 2016). The primer and probe sequences of this gene initially designed by Pang et al., (2004) were utilized in other studies after modifications (Freeman et al., 2008; Pang et al., 2011; Liu et al., 2011).
Real-time RT-PCR, a necessary tool to support the diagnosis and surveillance of rotavirus in Mexico
2018, Diagnostic Microbiology and Infectious DiseaseCitation Excerpt :Therefore, the original primers and probe; and the thermocycling conditions were modified; and additional primers were included in order to improve the sensitivity of the assay. Thus, the last modification in the original NSP3 protocol developed by Pang et al. it was able to detect 1 genome copy per reaction (Freeman et al., 2008; Mijatovic-Rustempasic et al., 2013; Pang et al., 2011; Zeng et al., 2008). The second real-time RT-PCR protocol to detect rotavirus from NSP3 gene was developed by Jothikumar et al. (2009), which it was able to detect all rotavirus genotypes and showed a limit of detection of 5 genome copies per reaction (Jothikumar et al., 2009).
